      【摘要】 目的 了解脑缺血肺损伤后肺组织中白细胞介素-1β(IL-1β)表达的变化,探讨其功能意义。 方法 成年SD大鼠随机分为假手术组和脑缺血肺损伤组,采用线栓法建立大脑中动脉脑缺血肺损伤模型。于术后24 h用RT-PCR(n=8),48 h用蛋白免疫印迹技术(n=8)检测肺组织IL-1β表达变化;术后72 h用免疫组织化学技术检测肺组织内IL-1β的定位分布(n=5)。结果 肺组织IL-1β基因和蛋白表达在脑缺血后24 h及48 h明显增加,与对照组比较,差异有统计学意义(P<0.05)。IL-1β主要分布在气道上皮细胞、巨噬细胞及中性粒细胞。结论 脑缺血肺损伤组织IL-1β表达明显上调,提示IL-1β可能在脑缺血肺损伤炎症反应中发挥作用。

      【Abstract】 Objective To study the expression changes of interleukin-1β (IL-1β) in the lung of rats with brain ischemia. Methods Adult SD rats were divided into sham operation group and brain ischemia lung injury (BILI) group randomly. Focal cerebral ischemia inflammatory lung injury model was developed with intraluminal thread technique. Lungs were harvested from rats at different time point respectively. RT-PCR (24 h), Western blot (48 h) and immunohistochemistry (72 h) were employed to detect the expressional changes and the distributions of IL-1β in the lung tissues. Results IL-1β immunohistochemical positive reaction was observed in epithelia cell and neutrophil as well as macrophage. Increased protein level and mRNA expression for IL-1β were found in lung after brain ischemia compared with those of sham group. Conclusion IL-1β, as a crucial inflammatory factor, could be associated with airway inflammation in lung following brain ischemia in rats.
